I just googled it and found what it was: It is a person who looked a the king's stools to see if they have grooves that would come from the stools "passing through something round protruding from the colon wall" i.e. they were checking to see if he had polyps.

Under Henry VIII, the Royal Court had a job title called "groove of the stool" for one of the personal servants of the king. Have a guess what this job really involved... :D
